"Eek, a Penis!" is the fifth episode of the twelfth season of South Park. It originally aired on April 9, 2008. In Canada, this episode aired on April 25, 2008.
This episode features Mrs. Garrison becoming male again for the first time since "Mr. Garrison's Fancy New Vagina".

Mrs. Garrison decides she wants to be a man again, and while she is gone, Cartman is put in her place as substitute teacher...

While their regular teacher, Ms. Garrison, is busy looking for a way to become the man he always intended to be, Cartman is put in charge. Soon after Cartman takes over, the kids' test scores are on the rise because of them cheating. Cartman decides to make a difference by working with some inner-city kids whom no other teacher has been able to reach by pretending to be a Spanish man and telling them to cheat on their tests. While teaching them how to cheat he tells them what to do if they get caught which is that they say "I Misinterpreted The Rules". One of his students doesn't want to cheat so leaves. Cartman meets up with this kid and plays Basketball with him, Cartman cheats by getting out a baton and hitting the student and scores twice, the student says he cheated but Cartman tells him "it doesn't matter just look on the scoreboard". Ms. Garrison is sobbing at home watching the news when it reveals that a biology research center have managed to grow human body parts on mice, he then goes there and asks them to create a penis on a mouse, they say they can't but Ms. Garrison pays them to. Ms. Garrison later comes back to check on her penis but accidentally lets it go so she goes running after it. He later runs into Stan, Kyle, Kenny & Butters who are playing in a sandpit he asks them "Boys have you seen my penis?" they reply "What!" he tells them that if they see it they should catch it. Then at butters' house his mom is washing the dishes and sees the penis and climbs up onto a stool or cardboard box and starts saying "eek, eek". All the students Cartman is teaching get 100% on their tests so are going to take a test in a small room being watched so they don't cheat but Cartman tells them that they should still cheat by teaching them how to make cheat sheet labels for their water bottles, they later get 100% on that test as well. Ms. Garrison is at the police station saying that his Penis is "missing", they get a call saying that a penis came into a Italian restaurant where Ms. Garrison chases the Penis into the spa where he pulls a naked man out by his penis. the penis runs away and jumps into a manhole. Ms. Garrison later is sobbing on a bench when his penis comes back to him. Then at the school they welcome back Cartman after teaching the inner-city students and Mr. Garrison after he became a man again and says that, "if you can't get pregnant, you're a dude!"

- The mouse with an ear on it's back is a reference to the Vacanti Mouse
- The song the mouse with the penis on its back sings in a reference to the film An American Tail.
- Cartman's stint teaching in an inner city school is a reference to the film Stand and Deliver.
- The high school Cartman teaches at is named Jim Davis High School. In Stand and Deliver, the school was Garfield High School. Jim Davis is the creator of the Garfield comic.
- The line when Principal Victoria is asking who wants to be a teacher and Cartman says me for several times, resembles the reminiscent scene from animated movie Shrek
